The Diamonds and Silver Ferns were locked in a typically tight battle for most of the game before the world champion Australian side produced a 15-8 final quarter to complete the 58-48 triumph.
The defence of captain Laura Geitz and Clare McMeniman turned into attack and the experienced Caitlin Bassett and Natalie Medhurst took advantage.
Medhurst, playing her 75th game for the Diamonds, landed 19 of her 20 shots and finished with 11 goal assists.
The third Test will be played in Melbourne on Sunday with the fourth and final game in Perth the following Friday.
